Project title:

Condition-dependent protease activation for targeted proteolysis in the regulation of DNA replication

Project is carried out within the Team 5/2017  programme of the Foundation for Polish Science

Project description:

The goal of this project is elucidation how, depending on conditions, proteases are structurally and spatially regulated to degrade essential replication proteins. The research will concentrate on bacterial Lon protease and its phosphate containing cofactors, contributing to the protease activities. By applying in vitro and in vivo experiments, including real-time, single-molecule and single-cell analysis with a variety of Lon proteins, substrates and cofactors, we expect to describe substrate-specific effects on Lon, and relate them to growth conditions. The cutting-edge techniques will be utilized for the description of the structure of Lon complexes. The research on molecular mechanisms of fundamental processes in bacteria is required due to the growing threat arising from the multidrug-resistant bacterial strains. Our goal is to provide data on Lon as a new target for new antimicrobial strategies based on the protease activity control. The project will be conducted with international collaboration of TEAM partners.

Key responsibilities include:

1.performing research using biochemical, genetic and molecular biology and computational techniques (independently and in collaboration with other team members)

2.participation in research design and data analysis

3.participation in preparation and writing of manuscripts

4.supervising younger researchers (Master students) involved in the project

5.active participation in lab meetings, departmental seminars and conferences

6.the project may require research stays in TEAM partners laboratories

Profile of candidates/requirements:

1.Master degree in one of the fields: biotechnology, biology, bioinformatics, chemistry

2.Solid laboratory training in biochemistry, molecular biology, microbial molecular genetics,

3.Training in bioinformatics, genomics, molecular phylogeny will be an additional advantage

4.Ability to communicate fluently in English (spoken and written)

5.Strong motivation to pursue scientific questions, creativity and analytical thinking
